Bacterial community structure of the marine diatom Haslea ostrearia ArchiMer

Haslea ostrearia produces a water-soluble, blue-green pigment, called marennine, with proven economic benefits (as a bioactive compound used to green oysters, which improves their market value). Incomplete knowledge of the ecological features of this marine diatom complicates its cultivation. More specifically, the ecology of bacteria surrounding H. ostrearia in ponds is what remains unknown. The structure of this bacterial community was previously analyzed by means of PCR-TTGE before and after isolating H. ostrearia cells recovered from 4 localities in order to distinguish the relative parts of the biotope and biocenose and to describe the temporal dynamics of the bacterial community structure at two time scales (2 weeks vs. 9 months). Bacterial community structure of the marine diatom Haslea ostrearia ArchiMer

The marine diatom Haslea ostrearia produces a water-soluble blue-pigment named marennine of economic interest (e.g. in aquaculture for the greening of oysters). Up to date the studies devoted to ecological conditions under which this microalga develops never took into account the bacterial-H. ostrearia relationships. COSELMAR, a 4-year project financed by the Région des Pays de la Loire at 2.1 M€, officially started on the 7th January 2013. The project is coordinated by Philipp Hess (IFREMER) and Sophie Pardo (Université de Nantes-LEMNA) and falls under the Federation for Research Institut Universitaire Mer et Littoral (IUML, FR CNRS 3473)*. COSELMAR has also been approved by the Scientific Council of the MSH Ange Guépin. La conchyliculture dans les pertuis charentais représente indéniablement l'une des activités économiques les plus importantes. Sur cette zone, ce sont près de 7800 ha qui sont occupés par des concessions ostréicoles, dont 3000 ha de marais littoraux (Prou et Héral, 1998). Ainsi le secteur de Marennes-Oléron représente, avec 25000 tonnes d'huîtres creuses produites en 1996, près de 18 % de la production nationale de ce mollusque, alors que ce secteur représente à lui seul près de 45 % de la commercialisation des huîtres à l'échelon national. First insight on interactions between bacteria and the marine diatom Haslea ostrearia : Algal growth and metabolomic fingerprinting ArchiMer

A first insight on the interactions between bacteria isolated from monospecific cultures of the marine diatom Haslea ostrearia and this microalgae is described in this work. From 28 bacterial isolates, the maximal growth rate and the maximal cell concentration were on average 58% and 27% higher (max. 67% for isolate B22). Isolates B5, B10 (both Saccharospirillum sp.) and B22 (Rhodobacter sp.) were the most efficient. The geographical origin of H. ostrearia from which bacteria were isolated had no impact on the algal growth performances, and a given bacterial isolate led to the same algal growth irrespective of the geographical origin of H. ostrearia. How essential are Argo observations to constrain a global ocean data assimilation system? ArchiMer

Observing system experiments (OSEs) are carried out over a 1-year period to quantify the impact of Argo observations on the Mercator Ocean 0.25° global ocean analysis and forecasting system. The reference simulation assimilates sea surface temperature (SST), SSALTO/DUACS (Segment Sol multi-missions dALTimetrie, d'orbitographie et de localisation précise/Data unification and Altimeter combination system) altimeter data and Argo and other in situ observations from the Coriolis data center. Two other simulations are carried out where all Argo and half of the Argo data are withheld. Les claires ostréicoles représentent près de 3000ha de marais exploités par la conchyliculture dans le seul Département de Charente-Maritime. Plus de 50000 tonnes sont ainsi affinées puis commercialisées à partir de ce bassin de production, représentant 1/3 de la commercialisation française. Les travaux menés en 1997 portent sur une meilleure connaissance écophysiologique des diatomées des claires ostréicoles dont Haslea ostrearia, ainsi qu'à de meilleures estimations des conditions environnementales naturelles (évolution de la qualité d'eau entre 2 remplissages de marais). En particulier, la variabilité et la répartition au sein des claires des populations phytoplanctoniques ainsi que l'évolution dans le temps de celles-ci ont fait l'objet de recherches....

Limiting nutrients of oyster pond seawater in the Marennes-Oleron region for Haslea ostrearia : applications to the mass production of the diatom in mesocosom experiments ArchiMer

Bioassays were carried out with the 'blue diatom' Haslea ostrearia Simonsen, which is responsible for oyster greening during the fattening period of Crassostrea gigas Thunberg in oyster ponds. Samples of seawater were taken from two oyster ponds: one without oysters and the other with 20 oysters per m_, maximal density allowed by the French AFNOR norm for 'refinement'. The aims were to clarify the nutrient requirements of this diatom, also to elucidate the eventual influence of C. gigas at this density on the seawater fertility and to envisage the mass production of this diatom by pond fertilization. Optimization of diatom Hasleao strearia cultivation in different mediums and nutrients ArchiMer

Hasleaostrearia is a marine diatom that can feed oysters. This study aimed to cultivate H.ostrearia in the following media: seawater (SW), seawater filtered by activated carbon (SW+CA), and drilling water (DW). The three-water media were supplemented with two types of nutrients, Walne-Conway+silica (CS) and ES-1/3 Provasoli (P), leading to a six experiment series. The cultures were grown in 200 mL and 400 mL respectively. The dilution cycle was every 3 - 4 day for 200 mL volume and every 4 - 5 days for the 400 mL volume for 20 - 40 days or until the cultures showed a decrease in growth. Seasonal variation in the antivibrio activity of two organic extracts from two red seaweed: Palmaria palmata and the introduced Grateloupia turuturu against the abalone pathogen Vibrio harveyi ArchiMer

The wide polarity range and highly polar compounds of two selected red seaweed, Grateloupia turuturu and Palmaria palmata were extracted using two different types of solvent, dichloromethane/methanol and methanol/water. Monthly in vitro antibacterial activities were studied using the microplate method against the marine bacteria Vibrio harveyi strain ORM4, known to infect abalone. Inhibition, slowdown and delay of Vibrio harveyi growth were investigated. Polar compounds of seaweed showed an activity against the abalone pathogen. The best activity was recorded from P. palmata collected in spring, with an inhibition of 7.9% of the bacterial growth. Preliminary 1H NMR profiles identified the differences between the extracts.

Cette étude constitue une des trois actions engagées dans le cadre du Contrat de Plan IFREMER - Poitou-Charentes (1994-1998) sous l"intitulé Valorisation des Claires Ostréicoles. Les deux actions complémentaires concernent la compréhension et l'amélioration du verdissement des claires sous la direction du Pr. J.M. Robert de l'Université de Nantes/lSOMER et la production de masse de microalgues afin de stimuler la croissance de mollusques filtreurs sous la direction de Dr. J. Hussenot du CREMA L'Houmeau.
